What MT-II is.

MT-II takes alpha-melanocyte-stimulating hormone and bends it into a ring: a lactam bridge locks the peptide's active core into a rigid cycle, producing a compact seven-residue molecule that is tougher, longer-lived and substantially more potent than the linear hormone.

The cyclization has a second consequence: where MT-I stays selective for the MC1 receptor, MT-II activates several members of the melanocortin receptor family — MC1R, MC3R, MC4R and MC5R. That breadth defines its research role.

Where the research points.

MT-II is the classic pan-melanocortin screening tool. Receptor-subtype comparison studies use it as the broad agonist against which selective ligands are profiled, and pigmentation research uses it for potent MC1R engagement in melanocyte and animal models.

Because MC3R and MC4R are central to energy-balance and appetite circuits in the brain, MT-II also appears widely in preclinical studies of those pathways — a reminder that the melanocortin system reaches well beyond pigment. It's equally a structural case study: the standard example of how cyclization changes a peptide's stability, potency and selectivity at once.
