What MOTS-c is.

MOTS-c is a sixteen-amino-acid peptide with a genuinely unusual birthplace: it is encoded inside the mitochondrial genome, in a short open reading frame hidden within the 12S ribosomal RNA gene. Its 2015 discovery helped establish a new class of molecules — mitochondrial-derived peptides — signals the cell's power plants send to the rest of the cell and body.

That origin upended a long assumption that mitochondrial DNA encodes only respiratory-chain machinery. MOTS-c showed the organelle also writes its own signalling molecules.

Where the research points.

The core literature is metabolic: MOTS-c has been reported to activate AMPK — the cell's low-fuel sensor — and to influence glucose handling and insulin sensitivity in rodent and cell models. Exercise physiology is the second major thread: muscle expression and circulating levels have been reported to respond to exercise, framing MOTS-c as a candidate 'exercise mimetic' signal under active investigation.

A third, striking finding drives newer work: under metabolic stress the peptide translocates to the nucleus and appears to act as a transcription-regulating signal — a mitochondrion reaching directly into nuclear gene expression. Retrograde signalling of that kind is one of the livelier corners of current mitochondrial biology.

It's encoded by the mitochondrion's own DNA rather than the nuclear genome. MOTS-c comes from a small open reading frame inside the mitochondrial 12S rRNA gene — part of a recently recognized family of such peptides that also includes humanin.

Because studies have reported that exercise raises its expression, and that administering it in rodent models reproduces some exercise-associated metabolic signatures, notably AMPK activation. Whether that framing fully holds is exactly what ongoing research is testing.
